Nitrofurans as radiosensitizers of hypoxic mammalian cells.

چکیده
Our studies showed that various nitrofuran derivatives have excellent radiosensitizing properties in hypoxic Chinese hamster cells at concentrations at which there is no effect on plating efficiency. The radiosensitizing effect is primarily a dose-modifying effect, with nitrofurantoin, nitrofurazone, and nifuroxime (each at 500 ¿M) in complete medium giving enhancement ratios of 1.65, 2.0, and 2.2, respectively. In parallel studies, the radiosensitizing effect of molecular oxygen (air-saturated conditions) was found to be dose modifying, with an enhancement ratio of ~2.9. Selective radiosensitization in hypoxia has been demonstrated in proliferating populations of cells in every phase of the cell cycle, as well as in nonproliferating populations which were contact inhibited at the time of irradiation. In all cell populations tested, these compounds did not alter radiosensitivity in air-saturated conditions. The permeability of phospholipid membranes to nitrofurazone in 0.1 M NaCl has been measured, and a permeability constant (K) of 1.5 (±0.7) X IO"5 cm/sec

Increased cell killing by metronidazole and nitrofurazone of hypoxic compared to aerobic mammalian cells.
Nitromidazole and nitrofuran derivatives comprise a large family of compounds, some of which have been shown to be hypoxic cell specific radiosensitizers in vivo and in vitro. The effects of metronidazole (2-methyl-5-nitroimidazole-1-ethanol) and nitrofurazone (5-nitro-2-furaldehyde semicarbazone) were studied on cell viability in vitro in the presence of air or nitrogen in the absence of radia...

As the molecular mechanisms behind tumor hypoxia have now been delineated, the important conclusion of the radiation oncologist Dr. Okuyama that “hypoxic milieu as the primary cancer environment” needs to be reexamined for its impact on current cancer diagnosis and treatment. It is apparent that, although hypoxic tumor cells cannot live without oxygen and nutrients, aggressive tumor cells isola...

We describe herein for the first time our medicinal electronomics bricolage design of hypoxiatargeting antineoplastic drugs and boron tracedrugs as newly emerging drug classes. A new area of antineoplastic drugs and treatments has recently focused on neoplastic cells of the tumor environment/microenvironment involving accessory cells. This tumor hypoxic environment is now considered as a major ...
متن کاملFluorescent nitroheterocycles for identifying hypoxic cells.
Binding of several nitroheterocycles by mammalian cells is a function of the ambient oxygen concentration; anoxic single cells bind up to 10 times as much of these drugs as do aerobic cells. We thus hypothesized that fluorescent nitroheterocyles could be used to quantitate the fraction of hypoxic cells in multicell systems, and this was tested in multicell spheroids using a relatively nontoxic ...
متن کاملRadiosensitizing hypoxic cells with new 3-nitro-1,2,4-triazole derivatives in vitro and in vivo.
The new regioisomer derivatives 4a-f and 5a-f of 3-nitro-1,2,4-triazole (3-NTR) were synthesized for the development of new radiosensitizers of hypoxic cancer cells for radiotherapy. N(2)-Substituted 3-NTR derivatives 5a-f were stronger radiosensitizers of hypoxic cells in vitro (Chinese hamster V79 cells) than N(1)-substituted 3-NTR derivatives 4a-f, but in vivo they were weaker (SCCVII carcin...
